A var iant o f the B16 me lanoma , clone C8471, which is non tumor igen ic in immunocompeten t mice, is capab le of i m m u n i z i n g such mice agains t the paren ta l , tumorigenic clone B~59 (1, 2). Clone B559 is not immunogen ic and does not induce concomi tan t t u m o r i m m u n i t y (3). T h e immunogen ic Ca471 var ian t was der ived from B559 cells by cont inuous growth in the t h y m i d i n e analog, 5 -b romodeoxyur id ine (BrdUrd) . 1 C3471 cells have undergone numerous pheno typ ic changes, inc lud ing loss of tumor igenic i ty , p igmen ta t i on and p lasminogen ac t iva tor act ivi ty , as well as induct ion of endogenous retrovirus (1, 4-6). O t h e r invest igators have also p roduced immunogen ic a l tera t ions of t u m o r cells by xenogeniza t ion with viruses (7-9), hap tens (10, 11), mutagens (12-15), and with an t ineoplas t ic drugs (16-19). The inab i l i ty o f Ca471 cells to form tumors reflects al terat ion(s) in in teract ions o f these cells wi th the host 's i m m u n e system, r a the r than an intr insic loss o f tumor igen ic po ten t ia l of C3471 cells (20). I m m u n o c o m p e t e n t mice moun t an i n f l a mma to ry response to C~471 cells wi th in 24 h af ter inocula t ion , and inhib i t t u m o r fo rmat ion by these cells (2, 21). Mice immunolog ica l ly compromised by t r ea tmen t wi th an t i -mouse thymocyte serum (ATS) or X i r rad ia t ion suppor t t u m o r fo rmat ion by Ca471 cells. In the exper iments repor ted here, we have mixed B559 cells wi th per i tonea l cells from C3471 ce l l immunized mice and inocula ted the mixtures into no rma l and immunosuppressed mice (Winntype assay) to ana lyze leukocyte popu la t ions responsible for i m m u n i t y to B559 melanomas . O u r results show tha t no rma l mice do not develop tumors when inocula ted with a mix ture of B559 cells and per i tonea l cells
